  <dc:description xml:lang="srp">mlečnih krava. Pretpostavlja se da je razvoj mastitisa povezan sa karakteristikama izolata S. aureus i ekspresijom kombinacije gena faktora virulencije. Da bi se problemi mastitisa mlečnih krava, koji su često perzistentni, sveli na najmanju moguću meru, poterbno je dobro poznavanje virulencije S. aureus, kako bi se na osnovu tih saznanja mogle da primene najefikasnije mere kontrole. Cilj ove disertacije je bio da se izvrši suptipizacija S. aureus izolovanih iz uzoraka mleka uzetih u slučajevima mastitisa krava. Za realizaciju postavljenog cilja definisani su zadaci da se: utvrdi prevalencija mastitisa krava izazvanih koagulaza pozitivnim stafilokokama u Mačvanskom i Kolubarskom okrugu (Republika Srbija); izvrši fenotipska i genotipska karakterizacija izolata koagulaza pozitivnih stafilokoka; ispita osetljivost koagulaza pozitivnih stafilokoka na antimikrobna sredstva i ispita prisustvo gena virulencije kod S. aureus izolovanih iz uzoraka mleka uzetih iz pojedinih četvrti vimena krava u slučajevima mastitisa.Tokom 20 meseci, primenom Kalifornija mastitis testa, ispitano je 4484 mlečnih krava, sa 288 farmi na području Mačvanskog i Kolubarskog okruga. Pozitivan mastitis test je utvrđen kod 1673 mlečne krave (37,3%). Staphylococcus aureus je izolovan na krvnom agaru. Na osnovu makromorfoloških, mikromorfoloških i biohemijskih osobina izolati poreklom iz uzoraka mleka krava pozitivnim na Kalifornija mastitis testu su identifikovani kao S. aureus. Kod svih izolata, koji su na osnovu fenotipskih osobina identifikovani kao S. aureus je primenom PCR metode dokazano prisustva nuc gena i 23S RNK gena, specifičnih za S. aureus. Svi izolati S. aureus su primenom disk difuzione metode po Kirbi Baueru ispitani na osetljivost na antimikrobna sredstva i prisustvo gena koji kodiraju glavne faktore virulencije (coa, clfA, cna, efb, fnbA, cap5, cap8, icaA, icaD, bap, mecA, pvl, sea, seb, sec, sed, see, ser, seg, seh, sei, sej, sep, tsst-1) primenom PCR.S. aureus, kao uzročnik mastitisa dokazan je kod 242 mlečne krave, pa je ukupna prevalencija mastitisa izazvanih ovim uzročnikom bila 5,4% (95% interval poverenja, CI=4,7–6,1%). Poveććan rizik od intramamarne infekcije S. aureus utvrđen je na malim porodičnim farmama (odnos verovatnoće, OR=4,2, 95% CI=2,6–6,6, P &lt; 0,001) i farmama srednje veličine (OR=3,5, 95% CI=2,2–5,7, P &lt; 0,001). Disk difuzionom metodom utvrđeno je da je najveći procenat izolata bio rezistentan na penicilin (74,7%), zatim na ampicilin (56,5%), kloksacilin (48,1 %), neomicin (42,8 %), gentamicin (27,0%), tetraciklin (14,7%), kombinaciju amoksicilina sa klavulanskom kiselinom (14%), linkomicin (12,6%), ceftriakson (10,9%), cefaleksin (8,8%) , a najmanji procenat izolata bio je rezistentan na sulfametoksazol- trimetoprim (4,9%). It is assumed that the development of mastitis is related to the characteristics of the S. aureus isolate and the expression of a combination of virulence factor genes. In order to minimize the problems of mastitis in dairy cows, which are often persistent, it is necessary to have a good knowledge of the virulence of S. aureus so that the most effective control measures can be applied based on this knowledge. This dissertation aimed to perform subtyping of S. aureus isolated from milk samples taken in cases of mastitis in cows. For the realization of the intent goal, the following tasks were defined: to determine the prevalence of cow mastitis caused by coagulase-positive staphylococci in Mačvanski and Kolubara districts (Republic of Serbia); to carry out the phenotypic and genotypic characterization of isolates of coagulase-positive staphylococci; to examine the sensitivity of coagulase-positive staphylococci to antimicrobial agents and to investigate the presence of virulence genes in S. aureus isolated from milk samples taken from individual quarters of udder cows in cases of mastitis. During 20 months, using the California mastitis test, 4,484 dairy cows were examined from 288 farms in the Mačva and Kolubara districts. A positive mastitis test was found in 1673 dairy cows (37.3%). Staphylococcus aureus was isolated on blood agar. Based on macromorphological, micromorphological, and biochemical characteristics, isolates originating from milk samples of cows positive for the California mastitis test were identified as S. aureus. In all isolates identified as S. aureus based on phenotypic characteristics, the presence of the nuc gene and 23S RNA gene, specific for S. aureus, was proven using the PCR method. All S. aureus isolates were tested for susceptibility to antimicrobial agents and the presence of genes encoding the main virulence factors (coa, clfA, cna, efb, fnbA, cap5, cap8, icaA, icaD, bap, mecA, pvl, sea, seb, sec, sed, see, ser, seg, seh, sei, sej, sep, tsst-1) using PCR.S. aureus, as the causative agent of mastitis, was confirmed in 242 dairy cows, so the overall prevalence of mastitis caused by this causative agent was 5.4% (95% confidence interval, CI=4.7–6.1%). An increased risk of S. aureus intramammary infection was found on small family farms (odds ratio, OR=4.2, 95% CI=2.6–6.6, P &lt; 0.001) and medium-sized farms (OR=3.5, 95% CI=2.2–5.7, P &lt; 0.001). By the disk diffusion method, it was determined that the highest percentage of isolates was resistant to penicillin (74.7%), followed by ampicillin (56.5%), cloxacillin (48.1%), neomycin (42.8%), gentamicin (27.0%), tetracycline (14.7%), the combination of amoxicillin with clavulanic acid (14%), lincomycin (12.6%), ceftriaxone (10.9%), cephalexin (8.8%), and the lowest percentage isolate was resistant to sulfamethoxazole-trimethoprim (4.9%).
